Depletion of<i>Prmt1</i>in Adipocytes Impairs Glucose Homeostasis in Diet-Induced Obesity

Protein arginine methyltransferase (PRMT) 1 is involved in the regulation of various metabolic pathways such as glucose metabolism liver and atrophy skeletal muscle. However, role PRMT1 fat tissues under disease state has not been elucidated to date. In this study, we delineate function protein adipocytes vivo. expression was abundant white adipose (WAT), which induced upon a high-fat diet mice by obesity humans. We found that adipocyte-specific depletion Prmt1 resulted decreased mass without overall changes body weight mice. Mechanistically, WAT led activation AMPK pathway, causal increased lipophagy, mitochondrial lipid catabolism, resultant reduction droplet size Interestingly, despite energy expenditure, observed promotion tissue inflammation an ectopic accumulation triglycerides peripheral knockout mice, promoted impaired insulin tolerance reminiscent mouse models lipodystrophy. These data collectively suggest prevents from excessive degradation limiting AMPK-mediated catabolism control whole-body homeostasis diet-induced conditions.
